Nerve agents are among the deadliest poisons known to man. In this issue, Wang et al. demonstrate the “dual-switch” function of TRPV4 in the treatment of nerve agent intoxication. TRPV4 is a key target of soman poisoning due to its modulation of NMDA-mediated excitotoxicity in the early stage of poisoning and NLRP3-medicated neuroinflammation in the late stage. As shown in this cover image, the brain is attacked by the nerve agent soman, and a defensive line formed by blocking TRPV4 and its downstream NMDAR and NLRP3 pathways can effectively protects the brain against the nerve agent.
