Yin et al. have revealed that 6 hours of acute sleep deprivation heightens pain sensitivity and suppresses the activity of medial habenula (MHb) cholinergic neurons. Chemogenetic activation of these neurons significantly mitigates sleep deprivation-induced hyperalgesia, likely through their projections to the paraventricular nucleus of the thalamus and the interpeduncular nucleus. The cover illustration draws inspiration from Chinese mythology, where Tang Sanzang’s recitation of the Tightening Spell, causing unbearable pain to Sun Wukong, symbolizes the heightened pain sensitivity resulting from acute sleep deprivation. In contrast, Guanyin Bodhisattva’s act of sprinkling sweet dew from her Jade Pure Bottle using a willow branch to relieve Sun Wukong’s suffering mirrors the therapeutic effect of activating MHb cholinergic neurons to alleviate sleep deprivation-induced hyperalgesia.
