Although neurons have been considered the primary producers of amyloid-β (Aβ), there is increasing evidence that non-neuronal cells—such as the myelinating glia of the central nervous system, oligodendrocytes—also contribute to Aβ production in Alzheimer’s disease (AD), which is highlighted in this review by Sasmita and Depp. Understanding how non-neuronal cells process and produce Aβ may reveal novel AD preventative/therapeutic strategies to stall Aβ accumulation from reaching threshold concentrations for Aβ plaque deposition, the primary hallmark of AD. The cover depicts an investigation board with key players in Aβ pathogenesis alongside other potential cellular contributors to AD pathophysiology, centrally highlighted and connected to other aspects of AD research that have spanned over 100 years.
