Although neuronal autonomous toxicity is considered the primary driver in the pathogenesis of Huntington’s disease, accumulating evidence suggests that oligodendrocyte-mediated white matter injury represents an early pathological event that cannot be overlooked. Li et al. demonstrated that exclusive expression of mutant huntingtin (mHTT) in oligodendrocytes is sufficient to induce robust activation of pro-inflammatory glial cells and the release of various inflammatory cytokines in the striatum and corpus callosum, raising the possibility that targeting non-neuronal mHTT toxicity may offer a novel therapeutic strategy. On the cover, the brain is depicted as a magical sea, where oligodendrocytes expressing mHTT act as the storm center, like a giant whale spouting water, disrupting the surrounding microenvironment and triggering pro-inflammatory changes in nearby astrocytes and microglia.
